As medical needs grew, the hospital transformed into a medical school in 1948. World War II had further underscored the importance of robust healthcare infrastructure, and the school catered to the medical needs of both the local population and allied troops stationed in the region. By 1962, the institution had blossomed into a full-fledged medical college, aptly named "Sylhet Medical College Hospital."

The college's affiliation with the prestigious Dhaka University in 1964 marked a significant milestone. With access to renowned faculty and advanced facilities, the quality of medical education soared. In 1978, the college shifted to its present spacious campus, offering a serene and conducive environment for learning and research.

To honor the contributions of General M.A.G. Osmani, a revered freedom fighter and chief of the armed forces during Bangladesh's Liberation War, the college was renamed "Sylhet MAG Osmani Medical College" in 1986.
